What a long wait for a reply! Hopefully you've found the way by now, but if not, here's how i go. I have only ever done the ride off peak.
Get yourself onto the Anzac Bridge, continue all the way. AT the bottom of the spiral ramp, cross and into Miller then straight all the way along Union until you're at the traffic lights looking at the Pyrmont Bridge. Cycle across this bridge, follow the bike/ped path and at the end go straight up King St cycle path. Then go left into Clarence or Kent (can't remember which). Turn Right off Kent and it will take you to the Harbour Bridge (use the footpath here). If it's peak hour, jsut follow the other cyclists
Down the Harbour bridge steps, look left to get you facing away from the tunnel. Go right at Alfred; at the roundabout go straight ahead into the Middlelmiss lane. Continue along (up) this lane all the way to the end. Left into Walker, then immediatelty right into McKenzie. Right into Miller. Left at Ernest, right onto West St,, at very end at T intersection go left into Palmer. Right into Hamilton, the bike/ped path starts at end of Quarry.
Ride on this path, follow the signs for Lane Cove and then Macquarie Uni/Park. Some of the signs are a bit hidden behind the bushes.
This will take you eventually to Epping Road. Follow the newish cycle/ped path all the way until Delhi Road. Cross at the lights. I get onto the city side footpath then cross Plassey Rd at the lights, ride past the crematorium. At the end of the road, ride into Lane Cove National Park. Left into Riverside Drive, then at the end (before the last 50metre uphill road for car exit) go straight ahead across a little bike/ped path with a cage fence. Follow this short steep path until you are on Lane Cove Road.
Right on the footpath, right onto Fontenoy (ride on the road from now), left into Khartoum. At this point, you can find the best way for you. On a weekend I go down Waterloo towards the uni, but if it was peak hour weekday, TAlavera road has a fair length of bike path and would be safer.
Here's a good way to get home,for some variety. Retrace your steps or else...go through Shrimptons creek bike path (off Waterloo Rd) all the way until Quarry Road. right at Aeolus to end. Leftish at end and cross Blaxland RD and first left all the way down Melville. Brake hard and right at Parkes, left at Hermitage.
Cross Victoria Road, straight ahead, around the back, and side of the Meadowbank TAFE. Left at the old house with a brass tap monument outside (angas ST?) and over the narrow bridge. First right, then left at the top into Railway Rd. There's a bike /ped access to John Whidddon bridge - its only about 10 m over the road and to the left. Follow this to the end, right into Blaxland.. AT the lights at the end, cross Concord Road, right for 100m then left onto path - ride through park, at end right into Killoola, left in Fremont, right into Hospital, left into Nullawarra.
I'm sure you can find your way to Stanmore from there. But if not, R Majors Bay, L Smythes, R Ian Parade, L Zoeller, Rt Tripod, L Burwood, Rt Ward or Marseau. Lyons Rd, then its up to you.

if you don't like that cross city route you can go left into Sussex from King St and then a few km's along Susses (at the Wharf theate) go right (ARgyle, Watson and Upper Fort) and ride up the hill to Observatory Hill). That's a much quieter way.
